По-видимому, самые фундаментальные вопросы клеточной биологии — это вопросы клеточной и тканевой архитектуры и такие связанные с ними явления, как подвижность клетки и митоз. Многие принципы, лежащие в основе сборки, функционирования и природы клеточных структур, начинают проясняться только сейчас. Относительно низкий уровень наших знаний в этой области можно объяснить, в частности, ограниченными возможностями тех методов исследования морфологических особенностей, которыми мы располагаем. Световой микроскоп позволяет выявить высокоупорядоченные субклеточные скелетные структуры, но обладает слишком малым разрешением. Что же касается стандартного метода тонких срезов в электронной микроскопии, то результаты, полученные с его помощью, весьма1 скудны, если не считать исследований клеточных структур, связанных с мембраной. Цитоскелет на обычном залитом срезе виден плохо, хотя из опытов с использованием флуоресцентных антител известно, что он существует. Ядра на стандартных срезах просматриваются лишь как скопления волокон хроматина, и, чтобы обнаружить более тонкую их организацию, приходится применять чрезвычайно сложные, специальные методики [36, 39].
